Advances in Supercritical Fluid Chromatography for the Analysis of Chiral and Achiral Pharmaceuticals
A revival of supercritical fluid chromatography (SFC) has been observed recently. SFC has repeatedly enabled fast and efficient separations, and in some cases has even outperformed high performance liquid chromatography (HPLC). This article provides an overview of the most recent advances in the field of chiral and achiral separations in SFC. This involves research focused on the most critical parameters in SFC separations, but also on practical issues such as the serial coupling of columns. The recent evolution from classic SFC to ultrahigh-performance supercritical fluid chromatography (UPSFC) is also discussed. More...
Automated MAb Workflow: From Harvest Cell Culture to Intact Mass Analysis of Variants
Recombinant monoclonal antibodies (MAbs) are a growing class of therapeutic molecules. Early in the development of MAbs, a large number of harvest cell culture (HCC) samples must be screened for immunoglobulin G (IgG) titer, aggregations, and charge variants. This study describes a two-dimensional (2D) high performance liquid chromatography (HPLC) workflow for affinity, size-exclusion, and ion-exchange analysis. More...
