Automated Ultrasonic Extraction and Filtration of a Plant Material
Ultrasonic extraction of glycosides was performed on plant materials for metabolomics studies. The extract was prepared for LC-MS/MS(QTOF) analysis by two filtration steps to prevent syringe blockage. Excellent reproducibility was obtained.

Fully automated multiple-heart-cutting analyses of blood plasma and induced sputum lipidome are demonstrated, using an Agilent 1290 Infinity 2D-LC solution coupled with an Agilent 6530 Accurate-Mass Q-TOF LC-MS system.

Increasing Efficiency of Method Validation for Metoclopramide HCl
This application note demonstrates the validation of a UPLC method for related substances of metoclopramide HCl for linearity, detection, and quantitation limits, accuracy, repeatability, intermediate precision, specificity, and robustness using Empower 3 Method Validation Manager. Validation results showed that the method is linear, accurate, repeatable, precise, specific, and robust for all related substances tested in this study. The quantitation limit of all related compounds was below the reporting threshold of 0.1% or 0.5 µg/mL.

LCMS Determination of Free Amino Acids in Beer
Beer is one of the most popular beverages worldwide. The composition of beer wort is largely dependent on the raw materials and the style of brewing used in the production. The contribution of specific amino-acids in the formation of off-flavors as well as the significance of amino acids as a source of nitrogen for fermentation is indisputable. Therefore, determination of changes in the composition and content of amino acids is an important measure in ensuring the consistency in quality of the end-product. In this study we used the Shimadzu UF-Amino station for automated pre-column derivatization and on-line reversed-phase LC separation with MS detection using an LCMS 2020 single quadrupole MS.

Extraction, Cleanup, and Concentration of Pesticides in Soil
Some pesticides have proven hazardous to other animals or the environment and have either been restricted or banned. The need to monitor food products for pesticides is essential, as more pesticides are being discovered to have adverse effects. One class of commonly used pesticides are chlorinated pesticides. These compounds are commonly detected using EPA Method 8081. This method determines the concentrations of various organochlorine pesticides in extracts from solid and liquid matrices, including food products, by GC with electron capture detectors (ECD). This new application speeds up and combines multiple sample prep processes into a single, automated method.

Quantitative Analysis of EtG and EtS in Urine Using FASi® ETG and LCâ€"MS-MS
Ethyl glucuronide (EtG) and ethyl sulfate (EtS) are conjugated ethanol metabolites formed following alcohol consumption. Analyzing chemical residues in biological matrixes requires pretreatment to eliminate non-desirable matrix components. Due to the polar nature of EtG/EtS many labs avoid pretreatment and use a dilute-and-shoot approach, leading to significant matrix suppression. UCT has developed a filter-and-shoot product for extraction of EtG/EtS from urine. Utilizing FASt® ETG with a Selectra® ETG HPLC column provides cleaner extracts and lower LOQ’s than achievable using a dilute-and-shoot method.

Extraction of Monohydroxy and Dihydroxy Vitamin D Metabolites from Serum
The method achieves high recoveries and is both sensitive and versatile enough to measure both the commonly measured 25 hydroxy metabolites and the better predicting 1,25 dihydroxy metabolites. The method involves SLE extraction followed by a simple PTAD derivatization.

Characteristics and Application of a Chiral Column, Chiral CD-Ph
"Chiral CD-Ph" is silica-based chiral packing material modified by phenyl-carbamated β- cyclodextrin (CD) as the enantiomer selector. Chiral CD-Ph is useful for the chiral separation of the racemic substance with benzene ring in its structure. It has superior capacity of chiral recognition derived from balanced hydrophobicity and polarity from the phenyl carbamate and the inclusion function from the central cavity in the structure of cyclodextrin. In addition, Chiral CD-Ph is applicable in both of the normal phase and reversed-phase mode, hence enable to provide separation conditions best matching the solubility of different samples. Chiral CD-Ph column is easy to use, with excellent durability.
