Rapid Sample Preparation for Determination of PAHs in Wild-Caught Avian Eggs Utilizing QuEChERS Extraction and Ostro 96-Well Plate Cleanup Followed by UPLC-UV Analysis

In this application note, a rapid and efficient method for the extraction of PAHs from a complex avian egg matrix was developed using established QuEChERS methodology and a one-step protein and phospholipid clean-up using Waters Ostro 96-well plates. This novel sample preparation approach eliminates the use of conventional GPC followed by SPE cleanup and successfully reduces sample preparation time to hours, instead of several days.  Download the app note
Multiresidue LC–MS–MS Screening of Veterinary Drugs in Milk Using the Ostro Sample Preparation Plate

This application note describes the use of the Ostro Plate for multiresidue screening for veterinary drugs in milk. Using this plate, milk samples are prepared using a few simple steps to effectively precipitate protein and remove potential interferences. As the extracted sample is passed through the plate the precipitated proteins are removed by filtration and phospholipid interferences are removed by retention to a proprietary sorbent. After this simple sample preparation, the reconstituted sample is analyzed using UPLC–MS–MS. To demonstrate the suitability of this method, representative compounds were chosen from major classes of veterinary drugs including tetracyclines, fluoroquinolones, sulfonamides, macrolides, beta-lactams, and beta-andrenergics. Download the app note

Analysis of Synthetic Cannabinoids from Whole Blood for Forensic Toxicology Using Ostro Sample Preparation Plates and CORTECS UPLC Solid-Core Columns

A panel of 22 synthetic drugs and metabolites were extracted fro whole blood and analyzed by UPLC–MS–MS. The use of Ostro Sample Preparation Plates resulted in excellent recoveries for all analytes with minimal matrix effects. Separation using the CORTECS UPLC Column enabled the analysis of all compounds in a short analysis time with baseline resolution of a critical isobaric pair. Download the app note
Direct Analysis of Opioids and Metabolites from Whole Blood Using Ostro Sample Preparation Plates Combined with UPLC–MS–MS for Forensic Toxicology

The method presented in this study demonstrates the use of Ostro Sample Preparation Plates combined with UPLC–MS–MS for the analysis of 22 opioid compounds and metabolites of interest in whole blood samples. All compounds were analyzed in less than 5.5 min with complete resolution of all isobaric compound pairs. The use of Ostro Sample Preparation Plates allowed for rapid, in-well cell lysis and protein precipitation, followed by elution into a 96-well collection plate. This procedure resulted in improved throughput compared to protein precipitation in individual tubes, with the added benefit of removing endogenous phospholipid compounds. All analytes demonstrated good linearity over the entire calibration range, and the method was sensitive for reliable detection at the lowest curve points. Download the app note

Extraction of Phospholipids from Plasma Using Ostro Sample Preparation

The Ostro 96-well Sample Preparation Plate is a pass-through device designed to capture and remove the highly abundant phospholipids as part of sample preparation for the analysis of small molecules during routine bioanalysis. In the generic protocol, the lipids are retained by the Ostro Pass-through Plate, and no attempt is made to release them. Modifying the loading and elution conditions allows this same affinity to be exploited for both selective enrichment of certain classes or for total phospholipid extraction. Download the app note
